Electron microscopic immunocytochemistry allows specific biological molecules to be localized in tissue sections at very high resolution. This book provides the information needed to use this technique. It covers the basic principles and gives examples of applications.
Electron microscopic immunocytochemistry is a powerful technique, combining the resolution of the electron microscope with the specificity and sensitivity of immunocytochemistry. Recent developments have simplified the techniques and broadened its range of applications, yet it is not used as widely as it could be, because it still has a reputation as an expensive and intricate procedure, beyond the grasp of most laboratories. This book aims to make the technique more accessible; it provides a thorough introduction to the principles of EM immunocytochemistry, detailed protocols, and examples of ways in which the technique can be applied to a variety of problems, in biomedical research and in diagnostic pathology. The emphasis throughout is on giving the practical help and guidance needed by scientists and clinicians who may be using the technique for the first time. The first part of the book provides an overview of EM immunocytochemical procedures and of applications in histopathology. Then three major chapters cover post-embedding, pre-embedding, and ultra-thin cryosection techniques.
The final section illustrates applications of EM immunocytochemistry to the study of endocrine tumours, renal disease, neuropathology, dermatology, and microbiology.
