This book deals with an exciting field of biomedical research: the application of recombinant DNA techniques to endocrinology. These and other new methods have made it possible to clone many endocrine genes, such as the insulin and growth hormone genes, to determine their nucleotide sequences, and to study their expression both in vitro and in vivo . The book covers the basics of gene cloning, structural analysis, and the regulation of gene expression in selected endocrine systems. The first part focuses on analytical methods and experimental approaches, including DNA-mediated gene transfer, identification of tissue-specific transcription factors and regulatory sequences, in situ hybridization of mRNA, and gene mapping on human chromosomes. The second part presents molecular studies of various endocrine systems, each combining different approaches. This book will be of interest to molecular biologists, biochemists and geneticists, as well as basic endocrinologists, since many endocrine genes were among the first eukaryotic DNA sequences to be isolated.

ANALYTICAL METHODS AND EXPERIMENTAL APPROACHES: M V Chao: Gene transfer and expression of mammalian cell receptors; A Dusty Miller: Transfer of endocrine genes using retrovirus vectors; C F Deschepper and others: In situ hybridization histochemistry on mRNA in endocrine tissues; Heng Wang and others: Factors required for transcription of the chicken ovalbumin gene; Michael D Walker and others: Cell-specific expression of pancreas genes: Role of 5' flanking sequences; Roger V Lebo: Mapping endocrine genes with sorted human chromosomes; MOLECULAR STUDIES OF ENDOCRINE SYSTEMS: Gary L Firestone: Glucocorticoid regulation of protein processing and compartmentalization; Walter L Miller: Structure, function, and hormonal regulation of steroidogenic enzyme genes; Michael Karin: Molecular analysis of steroid hormone action using the human metallothionein genes as a model; N L Eberhardt and others: Human growth hormone genes: Structure, evolution expression and hormonal regulation; William Chin: Hormonal regulation of glycoprotein hormone genes; J "org T Eppeln and Susumu Ohno: On DNA, RNA and sex determination; Graeme I Bell and Leslie B Rall: Mouse epidermal growth factor precursor: Sequence and expression in vivo of a multifunctional protein; Fu-Kuen Lin and others: Cloning and expression of human and monkey erythropoietin genes.
