Examines the chemistry, biochemistry, and biology of platelet activating factor (PAF) and considers the effect of existing anti-asthma drugs on responses to PAF to gain knowledge on the prospects for newer drugs in this field.
Platelet-activating factor (PAF) has recently been put forward as a mediator of asthma exacerbation, capable of iniating protracted cellular inflammation. This book examines the chemistry, biochemistry, and biology of this novel material and considers the effect of existing anti-asthma drugs on responses to PAF to gain knowledge on the prospects for newer drugs in this field.

Part 1 Platelet-activating factor; the discovery and present status of platelet-activating factor; biochemistry of platelet-activating factor; metabolism of platelet-activating factor; properties of platelet-activating factor (PAF) relevant to asthma; a platelet aggregation inhibitory factor in allergic patients; platelet-activating factor in normal rat uterine tissue; platelet-activating factor induced bronchoconstriction and extravasation in the guinea pig - effects of ketotifen and the PAF antagonist. Part 2 Inflammation and asthma: molecular and cellular inflammatory events and bronchial responsiveness; structural features and functional consequences of the inflammatory events in asthma; airway inflammation, airway hyper-responsiveness and exacerbation of asthma; a role for platelets in allergy and asthma; platelet-activating factor - relevance to asthma; specific binding by human eosinophils of the phospholipid mediator 1-0-alkyl-2-acetyl-sn-glycero-3-phosphocholine; effect of inhaled platelet-activating factor on bronchial responsiveness in non-asthmatic subjects; cellular, protein changes and leukotrienes in bronchial lavage fluid in patients with asthma; role of T lymphocytes in allergic inflammation of asthma. Part 3 Hyper-reactivity: the allergen-induced late asthmatic response and associated increases in nonallergic bronchial responsiveness; pharmacology of airway hyperreactivity in animals; the prophylaxis of asthma; enhanced IgE-dependent basophil histamine release and airway reactivity in asthma; bronchial airway hyperreactivity in a rabbit model for allergic asthma - response to histamine, acetylcholine, and platelet-activating factor; physiology of exercise induced asthma.
