This review summarizes current knowledge of the embryology of limb development and mechanisms that control the correct arrangement of limb structures. The review also highlights the progress that has been made in unravelling the molecular basis of pattern specification in developing limbs.
This review summarizes current knowledge of the embryology of limb development and mechanisms that control the correct arrangement of limb structures. These same mechanisms operate in both right and left limbs and thus it is difficult to account for the asymmetrical defects within this framework. The review also highlights the considerable progress that has been made in unravelling the molecular basis of pattern specification in developing limbs.

Experimental embryology of limb development. Treatment -induced congenital limb reduction defects (CLRD) in experimental mammals: classification of CLRD; classes of chemicals/treatment that causes CLRD; individual chemicals/treatment that consistently cause CLRD; thalidomide revisited; common mechanisms and relationship to normal limb development. Epidemiological studies of CLRD: background; literature search; classification of CLRD; prevalence at birth; possible specific causes of limb defects. Appendices: sample size for study; syndromes.
