This brief provides a broad overview of protein-engineering research, offering a glimpse of the most common experimental methods. It also presents various computational programs with applications that are widely used in directed evolution, computational and de novo protein design. Further, it sheds light on the advantages and pitfalls of existing methodologies and future perspectives of protein engineering techniques.

Dr. Krishna Mohan Poluri is currently working as an Asst. Professor at the Department of Biotechnology, IIT-Roorkee. He completed his PhD in Chemistry from the Tata Institute of Fundamental Research TIFR-Mumbai, and Post-doc from Rutgers University. Dr. Poluri worked as a Research scientist at the University of Texas Medical Branch (UTMB-Texas). His areas of expertise are Protein Engineering, Bio-molecular Spectroscopy, Structural Biology, Molecular Biophysics, Bio-molecular Interactions, Glycoimmunology, Structural virology and Structure based design of Therapeutics and Scaffolds. He is an editorial member and reviewer for several journals. He has won several awards and fellowships for his research work. Most prominent being: Young scientist awards from Indian Science Congress association (ISCA) -2009, National Academy of Sciences India (NASI)-2014, and Innovative Young Biotechnologist Award (IYBA)-2013 by DBT. Ms. Khushboo Gulati is currently pursuing her PhD at Department ofBiotechnology, IIT- Roorkee. She completed her Bachelors in Bioinformatics and Masters in Biotechnology from Punjab University, India. Her Research interests are in the area of Protein Engineering, Molecular Biophysics, Structural Biology and Glycoimmunology including Protein Carbohydrate interactions.
